New RNA Constructs Online


In the last few weeks the RNA production team has been very busy again and has made some new constructs. More than 100 RNA samples have been produced so far for NMR resonance assignments and fragment based screening.

In the first round all structural RNA elements located at the 5’ end, the ribosomal frameshift segment and the 3’-untranslated region (3’-UTR) of the SCoV2 genome were examined separately. In order to analyze stem-loop folding in the native sequence context 4 new constructs were selected and produced. Spectra of 5_SL1-4, 5_SL5, 3_SL1+2 and 3_HVR are now online, representing combinations of smaller stem-loops within their genomic nucleotide sequence.

Additionally, new spectra of RNA minimal motifs were recorded. These sub-constructs of the larger stem-loops 5_SL7, 5_SL8 and 3_SL1 werde used to confirm assignments and to get deeper insight into non-canonical structure regions. The imino proton assignments of 3_SL1sh_mut, 5_Sl7sh, 5_SL8loop and 5_SL8stem are now online.
